Enhancement of mangiferin in Aquilaria crassna Pierre ex Lecomte leaf extract

Abstract

Aquilaria crassna Pierre ex Lecomte belongs to the genus of Aquilaria, which is widely distributed in the tropical moist evergreen forests of Viet Nam. Mangiferin was purified and enhanced from Aquilaria crassna leaves by the adsorption – elution method using HPD100 adsorbent resin. The adsorption – elution process was investigated and selected alternately from a set of parameters in order to generate the highest mangiferin content: Sample solution with a concentration of 20 mg/mL was adsorbed with 3 g of HPD100 resin in 3 hours at room temperature. Then, the adsorbent resin was eluted twice with 60% ethanol, flow rate 2mL/min. The content of mangiferin in a powder product was determined by HPLC method with 14.768%, 7.5 times higher than that in dried leaf powder (1.97%). The product also has the significantly enhanced antioxidant capacity with an IC50 value of 19.43 ppm compared to the IC50 value of the extract of 29.92 ppm (using DPPH methods). Overall, the mangiferin refining process by adsorbent resin has demonstrated the ability to improve the content and quality of mangiferin in Aquilaria crassna leaves, promising the potential to apply in cosmetic and pharmaceutical products.
